Stephania Tetrandra extract

Kampo medicine, Stephania tetrandra Radix (Stephania) in Boi-ogi - increases insulin levels in blood and decreases blood glucose levels in streptozotocin (STZ) ddY -diabetischen mice. These effects of Stephania being membranaceus Astragalus Bunge Radix (Astragali) at Boi-ogi at (Liu et al., J. Med Traditional., 17, 253-260, 2000) high. In the present study, the effects of Stephania isolated alkaloids bis-benzylisoquinoline in hyperglycemia of diabetic STZ mice were studied. A major Benzylisochinoline-bis-alkaloid Fangchinolin (0.3 to 3 mg / kg), significantly decreased levels of diabetic glucose mice in the blood in a dose-dependent manner. The effect of Fangchinolin was 3.9 times higher than that of Stephania's water extract. Another major compound, tetranin (1-100 mg / kg), however, had no effect. The extract of Astragali water influenced not only but strengthened the anti-hyperglycemic effect of Fangchinolin (0.3 mg / kg). From compounds used (1 mg / kg) isolated from Stephania, Fangchinolin, Fangchinolin 2'-N-oxide and alpha-2'-N-Norfangchinolin which is substituted with a 7-hydroxy side chain 7 -O-methyl side are, Sank to nearly 50% high blood sugar. In addition, tetandrin-2'-N-beta-oxide, tetrandrin-2'-N-alpha-oxide, tetandrin-2-N-beta-oxide, Fangchinolin-2'-N-alpha-oxide, Or 2 are added 'side chain N-oxide, also refused to nearly 50% high blood sugar. Finally shows Fangchinolin but not Stephania tetandrine the anti-hyperglycemic effect of diabetic mice to STZ. Demethylation of the 7-O-position and / or addition of the side-chain 2- or 2'-N-oxide in the high-Benzylisochinoline Stephania connections have a role in inducing anti- Hyperglycaemic agents.
