Study the hormonal control of the expression of the PAI-1 gene and secretion in human adipose tissue in culture. It focuses on the effects of glucocorticoids, insulin, cAMP and catecholamines in explants of the omental region. The addition of dexamethasone in the culture medium increased the secretion of PAI-1 in a manner dependent of the time to 24 hours. Glucocorticoid stimulation was preceded by a 2-fold increase in mRNA levels between PAI1 4 to 8 hours of culture. The authors conclude that there is reciprocal regulation of PAI1 Dexamethasone (positive effector) and Camp / catecholamines (negative effectors) in human adipose tissue in culture. The stimulation by glucocorticoids may contribute to increased production of PAI1 high plasma levels of fat and PAI1 with central adiposity and thus a link between this disease and cardiovascular disease. Inhibition impaired by catecholamines may also contribute, as in vivo adipose tissue responses to these hormones are usually dull in obese people
