Isophorone diisocyanate Introduction

Isophorone diisocyanate (<99% purity) was purchased from Aldrich Chemical Corp., Milwaukee, WI (Lot # 02217JT) and was prepared in acetone / olive oil (4: 1), who also served vehicle. Primary irritation studies showed that 1.0% of minimal irritant concentration isophorone and treatment with 3.0% isophorone leads almost twice larger than 1.0% to a stimulus response. female B6C3F1 mice were sensitized by subcutaneous route at 0, 0.1%, 0.3% or 1.0% isophorone solutions daily for 5 consecutive days and 7 days later challenged with a solution of 3.0%. A 0.5% solution of 1-fluoro-2,4-dinitrobenzene (DNFB, Sigma Chem Corp.;. Lot No. 88F-3833;.> 99.0% pure as determined by HPLC) was used as positive control used. Site preparation included dermabrasion and intradermal injection of complete Freund's adjuvant in some mice. The mice were divided into groups of 8 to 10 mice / treatment group as shown in Table 1.
